The trial results, presented at the European Association for the Study of Diabetes meeting in Milan, highlight the efficacy of combining eloralintide—a selective amylin receptor agonist—with the dual GIP and GLP-1 receptor agonist tirzepatide. Over a 48-week period, the combination therapy led to a 23.3% reduction in body weight and a 2.9% drop in A1C levels. In comparison, patients treated with tirzepatide 15 mg saw a 14.8% weight reduction and a 2.4% decrease in A1C.

Eli Lilly's Triple-Action Drug Candidate Outperforms Tirzepatide
Participants in a Phase 2b trial for EloraTZP achieved an average weight loss of 54.1 lbs, significantly outpacing the 34.4 lbs recorded by those on tirzepatide alone. The experimental combination, which targets three hormonal pathways, also demonstrated superior blood sugar control in adults with obesity and type 2 diabetes.

Liana K. Billings, lead author and director of clinical research at Endeavor Health, noted that the data underscores the potential benefit of targeting multiple hormonal pathways simultaneously to manage the complex interplay between obesity and type 2 diabetes. While the combination showed higher rates of mild to moderate gastrointestinal side effects compared to individual therapies, Eli Lilly intends to move forward with the program. Based on these findings, the company plans to initiate Phase 3 trials for an optimized EloraTZP co-formulation by the end of 2026.
