The new candidate addresses a significant engineering challenge in nephrology. By targeting an unexplored region of the mRNA associated with the disease, XtalPi bypassed crowded patent landscapes that have historically limited therapeutic development for IgA nephropathy. In laboratory assays, the candidate achieved subnanomolar silencing potency, with an IC50 value tens of times lower than existing clinical-stage reference molecules.

XtalPi Advances AI-Designed siRNA Candidate for IgA Nephropathy
Nine months after launching its RNA research program, XtalPi has nominated a preclinical candidate for IgA nephropathy. The company credits its Kodexia™ platform, which merges generative AI with automated laboratory validation, for identifying a high-potency molecular design that outperformed clinical-stage reference molecules in early testing.

Testing in non-human primates revealed a durable pharmacodynamic effect, as a single dose maintained robust target-protein suppression for several months. This longevity suggests the potential for extended dosing intervals, a critical factor for patient compliance in managing chronic renal conditions. XtalPi’s workflow, which integrates first-principles modeling with continuous robotic experimentation, reportedly increased molecular design efficiency threefold compared to conventional industry methods. More than half of the initial designs produced by the platform have demonstrated superior in vivo activity compared to positive controls. The company now intends to advance this candidate toward clinical trials while exploring the platform's utility for multi-target siRNA designs and extra-hepatic delivery strategies.
